Thermo Scientific Taq DNA Polymerase, native, is a highly thermostable DNA polymerase of the thermophilic bacterium Thermus aquaticus. The enzyme catalyzes 5' to 3' synthesis of DNA, has no detectable 3' to 5' exonuclease (proofreading) activity, and possesses low 5' to 3' exonuclease activity. In addition, the polymerase exhibits deoxynucleotidyl transferase activity, which frequently results in the addition of extra adenines at the 3'-end of PCR products. Native Taq DNA Polymerase is preferred for amplification of bacterial DNA sequences homologous to those found in E. coli.
Besondere Eigenschaften
- Thermisch stabil Halbwertzeit beträgt bei 95 °C mehr als 40 min.
- Erzeugt PCR-Produkte mit 3'-dA-Überhängen.
- Lieferung mit zwei Puffern -10X Taq Puffer mit KCl und 10X Taq Puffer mit (NH4)2SO4. Letzteres ermöglicht eine PCR bei einem breiten Bereich von Magnesiumkonzentrationen und verringert unspezifische Grundierung.
- Inkorporiert modifizierte Nukleotide (z. B. Biotin-, Digoxigenin-, fluoreszierend markierte Nukleotide).
