Description

Zum Lysieren von Zellen unter nicht denaturierenden Bedingungen. Invitrogen™ ProcartaPlex™ Zelllysepuffer ist in 20 ml-Einheiten erhältlich, die gekühlt (für den häufigen Gebrauch) oder gefroren (für längere Zeiträume) gelagert werden sollten. Zusätzliche Proteasehemmer können problemlos hinzugefügt werden.

Zur Lyse adhärenter Zellen werden 400 μl Zelllysepuffer für eine 10 cm-Schale verwendet. Für nicht adhärente Zellen werden 400 μl Puffer pro 107 Zellen verwendet, nachdem sie in PBS gewaschen und pelletiert wurden. Den Puffer kühlen und PMSF unmittelbar vor der Verwendung zum Puffer hinzugeben.

Pufferzusammensetzung:

  • 20 mM TRIS-HCl (pH 7,5)
  • 150 mM NaCl
  • 1 mM Na2EDTA
  • 1 mM EGTA
  • 1 % Triton
  • 2,5 mM Natriumpyrophosphat
  • 1 mM β-Glycerophosphat
  • 1mM Na3VO4
  • 1 μg/ml Leupeptin
