The Mouse 4-1BB (CD137/TNFRSF9) ELISA quantitates Ms 4-1BB in mouse serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Ms 4-1BB. Principle of the method The Mouse 4-1BB solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. CD137 ist ein Mitglied der Superfamilie der TNF-Rezeptoren und trägt zur klonalen Expansion, zum Überleben und zur Entwicklung von T-Zellen bei. CD137 kommt sowohl als Monomer als auch als Dimer auf der Oberfläche von aktivierten T-Zellen vor. CD137 kann auch die Proliferation in peripheren Monozyten induzieren, die T-Zellapoptose durch TCR/CD3-getriggerte Aktivierung verbessern und die CD28-Kostimulation regulieren, um Th1-Zellreaktionen zu fördern. Die Expression von CD137 wird durch Lymphozytenaktivierung induziert. CD137 ist ein induzierbares, Rezeptor-ähnliches Protein auf der Zelloberfläche von aktivierten T-Zellen und Thymozyten der Milz. CD137 kommt sowohl als Monomer als auch als Dimer auf der Oberfläche von aktivierten T-Zellen vor.
