MAP2K6 (MKK6) S207E T211E was assayed in a two stage coupled reaction involving phosphorylation of MAPK14 inactive (PV3305) by MAP2K6 (MKK6) S207E T211E followed by phosphorylation of MBP substrate by activated MAPK14. The enzymes were diluted in enzyme dilution buffer before adding to the reaction. In stage one, MAP2K6 (MKK6) S207E T211E at 0.83μg/mL final concentration, is incubated with MAPK14 inactive at a final concentration of 35μg/mL in 50mM Tris (pH 7.5), 10mM MgCl2, 1mM EGTA, 2mM DTT, 0.01% Triton™ X-100, and 200μM ATP for 30 minutes at 30°C. In stage two, 5μL of the above reaction mix was transferred to 25μL of a reaction mixture containing 50mM Tris (pH 7.5), 10mM MgCl2, 1mM EGTA, 2mM DTT, 0.01% Triton™ X-100, 200μM ATP, 0.667μg/μL MBP substrate and trace [ 32P]-γ-ATP and incubated for 10 minutes at 30°C | |
