ELISA

The Mouse Interleukin-3 (Ms IL-3) ELISA quantitates Ms IL-3 in mouse serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Ms IL-3. Principle of the method The Mouse IL-3 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Rigorous validation Each manufactured lot of this ELISA kit is quality tested for criteria such as sensitivity, specificity, precision, and lot-to-lot consistency. Interleukin3 (IL-3), auch Mastzellwachstumsfaktor (MCGF), Multicolony Stimulating Factor (Multi-CSF) oder Eosinophil-CSF (E-CSF) genannt, wird von aktivierten T-Zellen, Mastzellen und Eosinophilen produziert. IL-3 ist ein hämatopoetischer Wachstumsfaktor, der die Koloniebildung von Erythroiden, Megakaryozyten, Neutrophilen, Eosinophilen, Basophilen, Mastzellen und monozytischen Linien stimuliert. Die meisten dieser Funktionen werden verbessert oder sind abhängig von der Kostimulation durch andere Zytokinen. Mensch- und Maus-IL-3 teilen sich nur 29 % Sequenzidentität auf Aminosäureebene. Die Bioaktivität von IL-3 ist sehr speziesspezifisch. Beispielsweise zeigt die menschliche IL-3-Zelle keine Aktivität bei Mauszellen. IL-3 fördert bei Zytokin starkes Wachstum. IL-3 ist in der Lage, die Proliferation einer breiten Palette hämatopoetischer Zelltypen zu unterstützen und ist an einer Vielzahl von Zellaktivitäten wie Zellwachstum, Differenzierung und Apoptose beteiligt. IL-3 besitzt nachweislich auch neurotrophische Aktivität und kann mit neurologischen Störungen in Verbindung gebracht werden.
