Viral Production Cells are a derivative of the HEK 293F cell line and are a core component of the LV-MAX Lentiviral Production System.

Gibco Viral Production Cells are a derivative of the HEK 293F cell line and are a core component of the LV-MAX Lentiviral Production System (Cat. No. A35684). Viral Production Cells have been adapted to a special chemically-defined, serum-free and protein-free LV-MAX Production Medium (A3583401). They are maintained in suspension culture and will grow to high density in LV-MAX Production Medium. Viral Production Cells are highly transfectable by LV-MAX Transfection reagent (A35348) and generate superior lentiviral production compared to polyethyleneimine (PEI)-based lentiviral production methods.

Gibco virale Produktionszellen sind ein klonales Derivat der HEK 293F-Zelllinie und ein Kernbestandteil des LV-MAX Lentiviral Produktionssystems. Virale Produktionszellen sind an ein spezielles chemisch definiertes, serumfreies und proteinfreies LV-MAX-Produktionsmedium angepasst. Sie werden in Suspensionskultur aufbewahrt und wachsen in LV-MAX-Produktionsmedium zu hoher Dichte an. Virale Produktionszellen sind durch LV-MAX-Transfektionsreagenz hochgradig transfizierbar und erzeugen im Vergleich zu Polyethylenimimin-basierten Methoden (PEI) der Lentivirus-Produktion eine höhere Ausbeute.

Gefrorene Zellen werden in einem Fläschchen mit 1 ml Zellen mit 1 x 107 lebensfähigen Zellen/ml in LV-MAX Produktionsmedium und 10 % DMSO geliefert. Die Zellen sollten direkt im LV-MAX-Produktionsmedium getaut werden.


Die Zellen werden in der Regel in Kolben auf einer Schüttelplattform in einem CO2-Inkubator mit hoher Luftfeuchtigkeit für Zellkulturen kultiviert. Die Kulturen sind jedoch im Volumen von 96 Deep-Well-Blöcken bis hin zu 2 l-Kolben und Bioreaktoren skalierbar. Die Lentivirus-Vektor-Ernte wird in der Regel zwei Tage nach der Transfektion durchgeführt.
