Gentamicin-Sulfat wird durch Fermentation von Micromonospora Purpurea oder M. echinospora hergestellt. Es handelt sich um eine Mischung aus 3 wichtigen Komponenten, die als C1, C1a und C2 bezeichnet werden. Jede Komponente besteht aus fünf Basis-Nitrogenen und benötigt fünf Äquivalente Schwefelsäure pro Mol Gentamicin-Basis. Gentamicin wird als Sulfat-Salz verwendet.

Gentamicin-Sulfat ist ein Breitbandantibiotikum, das gegen grampositive und gramnegative Bakterien wirksam ist. Es ist wirksam gegen Stämme, die beständig gegen Tetracyclin, Chloramphenicol, Kanamycin und Colistin sind, insbesondere Stämme von Pseudomonas, Proteus, Staphylococcus und Streptococcus.

Wirkungsweise:

  • Gentamicin verursacht Codon-Fehllesung durch Bindung an die ribosomale 30S-Untereinheit
  • Diese Bindung blockiert die Translokation von Peptidyl-tRNA von der Akzeptorstelle zur Spenderstelle
  • Die bakterizide Wirkung auf Pseudomonas aeruginosa wird durch die Bindung von Gentamicin an die äußere Membran ausgeübt, wo es natürliche Kationen verdrängt, die Membran destabilisiert und Löcher in der Zelloberfläche bildet.
