Description: This YCATE55.9 monoclonal antibody reacts with canine CD8 alpha. This cell surface receptor is expressed either as a heterodimer with the CD8 beta chain (CD8 alpha/beta) or as a homodimer (CD8 alpha/alpha). In dogs, CD8 alpha is expressed on thymocytes, a subset of mature T cells, NK cells, and intestinal intraepithelial lymphocytes (IELs). CD8 binds to MHC class I and plays a role in thymocyte development and mature T cell activation. This monoclonal antibody has been reported to inhibit cytotoxic T cell generation and function in vitro. In addition, this antibody has been reported to be immunosuppressive in vivo. Applications Reported: This YCATE55.9 antibody has been reported for use in flow cytometric analysis. Applications Tested: This YCATE55.9 antibody has been pre-titrated and tested by flow cytometric analysis of normal canine peripheral blood cells. This can be used at 5 μL (1.0 μg) per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. Super Bright 436 can be excited with the violet laser line (405 nm) and emits at 436 nm. We recommend using a 450/50 bandpass filter, or equivalent. Please make sure that your instrument is capable of detecting this fluorochrome.

When using two or more Super Bright dye-conjugated antibodies in a staining panel, it is recommended to use Super Bright Complete Staining Buffer (Product No. SB-4401) to minimize any non-specific polymer interactions. Please refer to the datasheet for Super Bright Staining Buffer for more information. Fixation: Samples can be stored in IC Fixation Buffer (Product No. 00-8222) (100 μL of cell sample + 100 μL of IC Fixation Buffer) or 1-step Fix/Lyse Solution (Product No. 00-5333) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation. Some generalizations regarding fluorophore performance after fixation can be made, but clone specific performance should be determined empirically. CD8 molecule is composed of two chains termed alpha and beta. CD8 is found on a T cell subset of normal cytotoxic / suppressor cells which make up approximately 20 to 35 % of human peripheral blood lymphocytes. The CD8 antigen is also detected on natural killer cells, 80% of thymocytes, on a subpopulation of 30% of peripheral blood null cells and 15 to 30% of bone marrow cells.
